Data has been obtained from studies conducted with Capa-305.
The acute toxicity of Capa-305 to zebra fish (B. rerio) in accordance with OECD 203, was based on nominal concentrations of 400, 200, 100, 50, 25, 12.5, 6.2 and 0 mg/l in ISO-water. The test solutions were renewed each day. Based on the total organic carbon content of the freshly prepared stock solution, the mean measured test concentrations were 319, 160, 80, 40, 20, 10, 5 and 0 mg/l. Based on measured concentrations, the 96h LC50 was 150 mg/l. The NOEC for mortality was 80 mg/l and the NOEC for effects on behaviour was 40 mg/L (Groenveld et al, 1992).
The toxicity of Capa 3050 to the water flea, Daphnia magna, was determined in a 48 hour static acute immobilisation test conducted according to OECD guideline 202. The daphnids were exposed to Capa 3050 at nominal loading rates of 28, 56, 113, 225, 450 and 900mg/l. Stability of Capa 3050 in the test media was confirmed by TOC analysis at the lowest and highest concentration at the start and end of test period. The 48 hour EC50 was >900 mg/l based on nominal loading rates (Helgesson, 2013).
Pseudokirchneriella subcapitata were exposed to Capa-305 at nominal loading rates of 28, 56, 113, 225, 450 and 900 mg/l in a static 72-hour static toxicity test carried out according to OECD 201 guidelines. The test design included an untreated control comprised of untreated algal growth medium only. Stability of Capa 3050 was confirmed in the test media.
The 72-hour NOEC based on inhibition of specific growth rate and inhibition of biomass was 113 mg/L. The 72 hour ErC50 was 490mg/l based on nominal exposure concentrations (Helgesson, 2013).
The toxicity of Capa-305 to the bacteria P. putida was tested in the cell multiplication inhibition test, at nominal concentrations of 0, 20, 40 and 80% of the saturated stock solution. No significant inhibition occurred at the highest test concentration during the 16 hour incubation period. The NOEC was 670 mg/L (Jansen & van den Berg, 1992).
In the chemical safety assessment performed according to Article 14(3) in connection with Annex I section 3 (Environmental Hazard Assessment) no hazard was identified. Therefore according to REACH Annex I (5.0) exposure estimation is not necessary for long-term toxicity to fish and aquatic invertebrates. Consequently, in accordance with Column 2 of REACH Annex IX, the study does not need to be conducted as all identified uses of the substance are assessed as safe for the environment.
