PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: 250 mg of the test substance was dissolved in a final volume of 1000 mL of dilution water to give a 250 mg/L stock solution. The pH was adjusted to pH ≥ 6 using 2M NaOH. Aliquots (22.4, 40, 72, 128, 224 and 400 mL) of the stock solution were each separately diluted in a final volume of 10 L of dilution water to give the final concentrations.

TEST ORGANISM
- Common name: rainbow trout
- Method of holding: fish were held in a temperature controlled room under artificial light, with a 16-h light: 8-h dark photo-period, in holding tanks under continuous water renewal (flow-through) conditions. The water used for the holding of fish stocks and for the toxicity tests was laboratory mains supply. The water was pumped to the laboratory through an activated carbon filter.

ACCLIMATION
- Acclimation period: at least 12 d prior to testing
- Type and amount of food during acclimation: fish were fed with a proprietary fish food, which was added to the holding tank in quantities dictated by the size of the fish. Uneaten food and debris was siphoned or cleaned from the tanks as required.

ANALYTICAL RESULTS

Analysis of the freshly prepared media resulted in measured concentrations of 70% to 105% of nominal value. Analysis of the corresponding old media resulted in measured concentrations of 72 to 101% of nominal value with the exception of the 1.0 mg/L test concentration at 96 hours which resulted in a measured concentration of 58% of nominal.

The temperature did vary by more than 1ºC in some instances. This deviation was considered not to affect the integrity of the study given that no adverse effects were observed in the control groups and the temperature remained within the 15 ± 2ºC range throughout the tests.

Additional information

One study is available for the substance hydroxylammonium chloride (CAS 5470-11-1), in which the short term toxicity to fish is tested. The study was conducted according to OECD guideline 203 and GLP. The test organism, Oncorhynchus mykiss was exposed in a semi-static freshwater system to nominal test item concentrations of 0.56, 1.0, 1.8, 3.2, 5.6 and 10 mg/L (geom. mean meas.: 0.484, 0.834, 1.50, 2.48, 5.03 and 10.3 mg/L) for 96 h. The test media was renewed daily during the test. The test concentrations were monitored at time points 0 (fresh media), 24, 48, 72 (old and fresh media) and 96 h (old media) by LC-TOF/MS. The measured concentrations ranged between 58 and 105% of the nominal values. Therefore the effect values were expressed as geom. mean meas. concentrations. The 96 h LC50 was determined at 1.78 mg/L (geom. mean meas.).
