Genetic toxicity in vitro

Description of key information

The substance benzol forerunnings (coal) contains > 0.1% Benzene and can also contain > 0.1% 1,3 -butadiene. Benzene is a known mutagen.

Benzene is known to show mixed results in an in vitro test, in some in vitro tests however benzene shows a clearly positive response ( bacterial mutation, mammalian cell gene mutation and mammalian cell chromosomal damage. In vivo testing in mammalian cells has shown potential for mutagenic toxicity and found positive results for both somatic and germ cells. Based on all these results benzene is considered positive for genotoxicity.

Also 1,3-butadiene has shown positive results in mutagenicity testing, resulting in a cat 1B mutagenicity harmonised classification.

Due to the significant presence of benzene and 1,3 -butadiene in benzol forerunnings (coal) the substance as a whole also has to be considered as a mutagen.

Justification for classification or non-classification

CAS 65996 -88 -5 has a harmonised classification as H340 (Muta. 1B).

The mutagenicity classification is in addition also supported by the presence of benzene and 1,3 -butadiene, known genotoxic carcinogens, in quantities in excess of 0.1%. Both of those substances also have a harmonised classification as a mutagen.
