PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
Method- Since ASA404 C7 was hardly soluble in water test concentrations were prepared separately in Milli-RO water (tap water purified by reverse osmosis; Millipore Corp., Bedford, Mass., USA). One litre test bottles were filled with 250 ml of test substance mixtures with initial loading rates twice as high as the final test concentration. These mixtures were stirred in closed bottles for 25-27 hours. Subsequently, 16 ml synthetic sewage feed, 200 ml sludge and Milli-RO water up to 500 ml were added resulting in the final loading rates (see also section 6.4). Optimal contact between the test substance and test medium was ensured applying continuous aeration and stirring.

The sludge was coarsely sieved, washed and diluted with ISO-medium. A small amount of the sludge was weighed and dried overnight at ca. 105°C to determine the amount of suspended solids (3.8 and 4.4 g/l of sludge, as used for experiment 1 and 2, respectively). The pH was 7.8 on the day of testing in both experiments. The batch of sludge was used one day after collection; therefore 50 ml of synthetic sewage feed was added per litre of activated sludge at the end of the collection day. The sludge was kept aerated at test temperature until use.

In the first experiment 62 and 69% inhibition of the respiration rate of the sludge was recorded at a loading rate of 1000 mg ASA404 C7 per litre.

In the second experiment no significant inhibition of the respiration rate of the sludge was recorded up to a loading rate of 180 mg/l (based on nominal concentrations). At a loading rate of 320 and 560 mg/l very slight inhibition (13%) of respiration rate of the sludge was recorded. Whereas, 80% inhibition of the respiration rate of the sludge was recorded at a loading rate of 1000 mg/l.

The EC50 was estimated to be at a loading rate of 770 mg/l (based on nominal concentrations).
