PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: An amount of test material (2000 mg) was added to the culture medium and stirred for 23 h to give the 100 mg/L loading rate. The stirring was stopped after 23 h and the mixture allowed to stand for 1 h. The water accommodated fraction (WAF) was then removed by filtration through 0.45 µm filters to give the 100 mg/L loading rate WAF. An aliquot (2 L) of the 100 mg/L loading rate WAF was mixed with algal suspension (20 mL).
- Controls: The control group was maintained under identical conditions but not exposed to the test material.

The test exhibited an EL50 of > 100 mg/L (nominal) based on the growth rate. No effects were observed in the range of the water solubility of the test substance. Chemical analysis of the 100 mg/L loading rate WSFs at 0 and 72 h showed the measured test concentrations to be less than the limit of quantification of the analytical method (0.18 mg/L). Exceeding the duration of stirring did not result in significant increase in TOC in test solutions.

Justification for type of information:
In accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5 a read across to the structurally similar analogue substance Dipentaerythritol with fatty acids C5 and C9 iso (CAS 647028-25-9) was conducted to fulfil the data requirements according to Annex VII of Dipentaerythritol hexaesters of 3,5,5-trimethylhexanoic and n-heptanoic acids. (CAS 1379424-11-9) in regard to toxicity to aquatic algae.
This read-across is justified in detail in the overall summary (IUCLID section 6.1) and within the analogue justification in IUCLID section 13. The source substance consists of the same alcohol component (dipentaerythritol) and similar fatty acid chain lengths as the target substance (C7, C9-iso for the target substance compared to C5, C9-iso for the source substance).

The study with the read across substance Dipentaerythritol with fatty acids C5 and C9iso investigated the toxicity to algae in a GLP guidance study. The test was performed according to OECD 201 (Mead and Mullee, 2000) with Scenedesmus subspicatus exposed to a water accommodated fraction with a limit concentration of 100 mg/L. The test substance concentration was analysed using GC-analysis and resulted in a concentration of <0.8 mg/L (limit of quantification). No inhibition was observed after 72 h. Therefore, this study resulted in an EL50 of > 100 mg/L (nominal). The NOELR was derived to be 100 mg/L (nominal).

Based on the result from the structurally related substance, in accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5, characterized by a similar ecotoxicological profile, it can be concluded that Dipentaerythritol hexaesters of 3,5,5-trimethylhexanoic and n-heptanoic acids will not exhibit toxic effects to aquatic algae up to the limit of water solubility.
