Short-term toxicity

Additional information
The evaluation of the short-term toxicity is evaluated by considering data both on the substance itself and on the similar substances. Justification for Read Across is given in Section 13 of IUCLID.
At least one study for any of the three acute trophic levels was reported with EC50 higher than 100 mg/l and showing general no toxicity at high doses.
Regarding long term studies, a Daphnia magna reproductive study is available on the substance.
A reproduction test on Daphnia magna (semi-static, 21 days) was carried out to assess the effect of 5 -A on the reproductive output of Daphnia magna, according to the OECD Guideline 211. Ten daphnids, held individually, were used per concentration level and control. Nominal concentrations of the test item were selected based on the results of a preliminary range finding test (non-GLP) as follows: 0.429 - 1.20 - 3.36 - 9.41 - 26.3 mg/l corresponding to the time-weighted mean measured test item concentrations were: 0.398 – 1.12 –3.04 – 8.48 – 23.4 mg/l. The overall effect threshold for effects of the test item on daphnid reproduction under the test conditions was 3.04 mg/l(NOEC) and 8.48 mg/l (LOEC) based on the nominal concentrations of the test item.
The effects of 5 -A on the early-life stage of fish (Danio rerio / Zebrafish) were determined at the test facility according to OECD Guideline 210. Based on the results of a range finding test the test was conducted as an extended Limit test with the nominal test item concentrations 1.25 and 12.5 mg/l. The test item partly caused significant effects on Zebrafish in an early life stage test, 30 days post hatch when tested with nominal concentrations of 1.25 and 12.5 mg/l. For the parameters hatch, post-hatch survival and overall survival the NOEC was 12.5 mg/l. Therefore, the respective LOECs were determined to be > 12.5 mg/l. For the parameters fry growth (expressed as fresh weight and length) the NOEC was < 1.25 mg/l. Therefore, the LOEC for these parameters was determined to be 1.25 mg/l. All effect values are given based on the nominal concentrations of the test item.
The effects of on the early-life stage of fish (Danio rerio / Zebrafish) were also evaluated for the similar substance 2 -A, determined at the test facility according to OECD Guideline 210. Based on the results of a range finding test the test was conducted as a dose-response test with the nominal test item concentrations 0.625, 1.25, 2.50, 5.00 and 10.0 mg/l. The test item caused significant effects on Zebrafish in an early life stage test, 30 days post hatch. For the parameter hatch, the NOEC was 10.0 mg/l. Therefore, the respective LOEC was determined to be > 10.0 mg/l. For the parameters post hatch survival and overall survival, the NOECs were 1.25 and 2.50 mg/l, respectively. Therefore, the respective LOECs were determined to be 2.50 and 5.00 mg/l. For the parameter fry growth (expressed as length and fresh weight) the NOECs were < 0.625 mg/l (length) and 0.625 mg/l (weight). Therefore, the LOECs for length and weight were determined to be 0.625 and 1.25 mg/l, respectively.
Considering the available data on the three trophic levels, fish is considered to be the most sensitive species as the effect level during the fish early-life stage toxicity study on 2 -A was the lowest obtained.
Therefore, SELECTED NOEC for aquatic toxicity: 0.625 mg/l from the fish early-life stage toxicity on the similar substance 2 -A.
