GC-MS method
Details on sampling:
- Sampling method: All concentration levels and the control were analytically verified via GC-MS after 0 (start of exposure) and 72 hours (end of the exposure). Separate replicates for each test item concentration and control for the test item analysis at the beginning of the exposure were prepared with algae. The samples for the test item analysis after 72 hours were prepared with algae and incubated under test conditions.
- Sample storage conditions before analysis: all samples were stored at 6 ± 2 °C until sample preparation andat room temperature until the start of the analysis (on an autosampler), if necessary.
Vehicle:
no
Details on test solutions:
A saturated solution with a concentration of 1 mL/L test item was prepared with dilution water. A slow stirring procedure was applied. Gentle stirring (to avoid formation of an emulsion) was carried out with a magnetic stirrer at room temperature for 72 hours. After a separation phase of 15 minutes at room temperature the saturated solution was taken from the bottom of the flask and was used for testing. The solution was checked after stirring via laser beam (Tyndall effect) for undissolved test item (formation of an emulsion). The Tyndall effect was negative.

Conclusions:
The growth inhibition effect of the test item on green algae was determined. The EC50-values for inhibition of growth rate (ErC50) and yield (EyC50) after 72 hours were 470 µg/L and 122 µg/L, respectively. The EC10-values for inhibition of growth rate (ErC10) and yield (EyC10) after 72 hours were 119 µg/L and 25.3 µg/L, respectively. The NOEC-values for both inhibition of growth rate and yield after 72 hours were 28.4 µg/L.
Executive summary:

The growth inhibition effect of the test item on green algae was determined according to OECD TG 201 and GLP. The test was conducted in a closed static system without headspace. Based on the results from the range-finding test, five concentrations with a spacing factor of 3.16 and one control were tested. The test concentrations were verified via GC-MS method. At the start of the exposure the measured concentrations were 9.55 - 32.8 – 93.5 - 300 – 827 µg/L and after 72 hours the amounts were between 75 and 103% of the initial concentrations. The geometric mean measured test item concentrations were calculated to be 8.69 – 28.4 – 84.8 – 269 – 839 µg/L. Therefore, the geometric mean measured concentrations were applied to calculate the biological effect of the test item. Under the test conditions, the EC50-values for inhibition of growth rate (ErC50) and yield (EyC50) after 72 hours were determine to be 470 µg/L and 122 µg/L, respectively. The EC10-values for inhibition of growth rate (ErC10) and yield (EyC10) after 72 hours were 119 µg/L and 25.3 µg/L, respectively. The NOEC-values for both inhibition of growth rate and yield after 72 hours were 28.4 µg/L.

Description of key information

A study on the growth inhibition effect of the item to green algae is available. The test was conducted in a closed static system without headspace according to OECD TG 201 and GLP.
