Description of key information
Three valid tests have been reported for the endpoint. In a 1996 OECD 209 study with activated sludge an EC 20 (30 min) > 1000 mg/L was reported. In a 1989 ISO 8192 B study an EC 20 (30 min) of 1000 mg/L was observed. A guideline-conform study of Butandioldivinylether on the celI multiplication of the bacterium Pseudomonas putida from 1996 resulted in an EC10 (16 h) of 5650 mg/L. The 1996 OECD 209 study was selected as key study and its result to be used as key value for chemical safety assessment, as this study was conducted with activated sludge from a wastewater treatment facility, thus reflecting more realistically to the real environmental setting when compared to the study with Pseudomona putida.

Key study BASF 1996
The inhibitory effect of Butandioldivinylether on activated sludge was investigated in a 30-min static test according to OECD Guideline 209 and ISO 8192. The sludge was taken from a laboratory scale wastewater treatment plant which was operated with domestic and synthetic wastewater. The influence of the test substance on the sludge respiration compared to the control was determined. The test substance was tested at the concentration of 1000 mg/L. An EC20 > 1000 mg/L was reported (the actually measured repiratory inhibition was 17%). The EC50 was slightly exceeded for the reference substance. Nonetheless the quality of the sludge was considered as good by the study director, and thus the test result as valid. The study was selected as key study and its result to be used for chemical safety assessment, as this study was conducted with activated sludge from a wastewater treatment facility, thus reflecting more realistically to the real environmental setting when compared to the study with Pseudomona putida.

The inhibitory effect of Butandioldivinylether on the celI multiplication of the bacterium Pseudomonas putida was investigated in a 16-hour static test according to DIN EN IS0 10712 (February 1996). The bacteria are cultured under specified conditions and the influence of the test substance on the cell multiplication compared to the control population is determined. After the incubation time the optical density of the bacterial suspension is measured in a photometer at a wavelength of 436 nm. The measured extinctions of the treated samples are compared to the untreated control to show possible toxic effects. The test substance was tested in the concentration range between 39.06 and 10000 mg/L (nominal). The dilution factor was 2.
The following results show the nominal concentrations of the test sample that causes after 16 h an of inhibition bacterial celI multiplication by 10% (EC10), 50% (EC50) and 90% (EC90) compared to the untreated control:
EC10 (16 h) = 5650 mg/L (nominal concentration)
EC50 (16 h) > 10000 mg/L (nominal concentration)
EC90 (16 h) > 10000 mg/L (nominal concentration)
The validity criteria were fulfilled by the test.
Supporting study BASF 1989
The inhibitory effect of Butandiol-1,4-divinylether on activated sludge was investigated in a 30-min static test according to ISO 8192. The sludge was taken from a domestic wastewater treatment plant and the influence of the test substance on the sludge respiration compared to the control was determined. The test substance was tested in the concentrations 466.4 and 1019.2 mg/L were tested (dilution factor ca. 2.2). An EC20 of 1000 mg/L was reported. No reference substance was used for control of the sludge sensitivity against respiratory inhibiting substances.
