In a bioaccumulation study, corresponding to OECD Guideline 305C, a BCF of <0.5 - 15 L/kg at 0.2 mg/L test concentration and <4.6 - 11 L/g at 0.02 mg/L test concentration for menthol was determined with Oryzias latipes as test organism within 6 weeks.

Description of key information
The bioaccumulation of menthols in fish (Cyprinus carpio) was investigated in a test according to standard method from Japanese competent authority. Experimentally determined BCF values in the range of <0.5-15 L/kg indicate no significant bioaccumulation potential of menthols.

Additional information
Since no data on the bioaccumulation propriety of the target substance are available, the potential for bioaccumulation is assessed on the basis of data from the structurally related menthol (stereo isomers). This read-across is in accordance with Regulation (EC) No 1907/2006 Annex XI, 1.5. The target substance is a racemic mixture of D-, and L-menthol. This allows the use of experimental data from another isomeric mixture of D-menthol and L-menthol for the assessment of the target substance. A detailed category approach justification is provided in the technical dossier (see IUCLID Section 13).
The study with the source substance was conducted according to OECD 305C with freshwater fish (Cyprinus carpio). Fish were exposed to two different concentrations (0.2 and 0.02 mg/L) for up to 6 weeks. BCF values were determined in the range of 0.5 - 15 L/kg (0.2 mg/L) and 4.6 - 11 L/kg (0.02 mg/L). Both BCF values do not indicate a significant bioaccumulation potential. The slight variation of BCF values can be explained by the variation of the lipid content in fish (2-6%).
Based on the result from one structurally related source substance (in accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5) it can be concluded that testing the target substance would result in comparable results.
