Description of key information
Short-term EC50 and NOEC values for 1,4-Dithiothreitol (CAS no. 3483-12-3) are available for two trophic levels of the aquatic freshwater compartment (aquatic invertebrates, algae) from GLP studies according to OECD 201 and 202.
Growth inhibition study aquatic algae (Raphidocelis subcapitata) (OECD 201):
ErC50 (72h): 24.3 mg/l nominal concentration
NOErC (72h): 3.2 mg/l nominal concentration
Short term toxicity on invertebrates (Daphnia magna) (OECD 202):
EC50 (48h): 34.8 mg/l nominal concentration
NOEC (48h): 25 mg/l nominal concentration

Algae:
The acute toxicity of 1,4-Dithiothreitol (CAS no. 3483-12-3) to the green alga Raphidocelis subcapitata was determined in a 72 hour static test according to OECD guideline 201. The median effect concentrations (EC50) of 1,4-Dithiothreitol on the green alga Raphidocelis subcapitata was estimated to be 24.3 mg/l nominal concentration after 72 h of exposure. The NOEC values were determined to be 81.3 mg/l nominal concentration after 72 h of exposure. The results of the test can be considered reliable without restriction. The study was rated Klimisch 1.
Daphnia:
The acute toxicity of 1,4-Dithiothreitol (CAS no. 3483-12-3) to Daphnia magna was determined in a 48 hour semi-static test according to OECD guideline 202. The median effect concentrations (EC50) of 1,4-Dithiothreitol on Daphnia magna were estimated to be 59.8 and 34.8 mg/l nominal concentration after 24 and 48 h of exposure, respectively. The NOEC values were determined to be 25 mg/l nominal concentration after 24 and 48 h of exposure, respectively. The results of the test can be considered reliable without restriction. The study was rated Klimisch 1.
Conclusion on classification
The environmental classification of 1,4-Dithiothreitol (CAS no. 3483-12-3) is currently not harmonised according to the CLP Regulation (EC) 1272/2008. Based on available acute aquatic toxicity data, the lowest valid effect concentration for freshwater aquatic organisms is 24.3 mg/L (EC50 for algae). Based on the available acute data, the substance should not be classified for acute toxicity following Annex I, table 4.1.0a of the CLP regulation (there are no acute values <=1 mg/L).
