Preparation of test solutions started with a loading rate of 100 mg/L applying a three-day period of magnetic stirring to ensure maximum dissolution of the test item in medium. Thereafter, the aqueous Saturated Solution (SS) was collected by means of filtration through a 0.45 μm membrane filter (RC55, Whatman) and used as the highest test concentration. Lower test concentrations were prepared by subsequent dilutions of the SS in test medium. All test solutions were clear and colorless at the end of the preparation procedure. Any residual volumes were discarded.

Executive summary:

A fish short-term toxicity test was performed according to OECD guideline No. 203 and in compliance with GLP criteria. In addition, procedures were based on the test methods described in the OECD series on testing and assessment number 23, 2000.

Seven fish per group were exposed to a control and to 4.6, 10, 22, 46 and 100% of the SS for 96 hours under semi-static conditions (daily renewal). Samples for analytical confirmation of exposure concentrations were taken at the start and at the end of the first and the last renewal period. At the start of the first renewal period, the actual measured test concentrations were 0.017, 0.037, 0.10, 0.22 and 0.46 mg/L in 4.6, 10, 22, 46 and 100% of the SS, respectively. At the end of this 24-hour renewal period these concentrations had decreased to 34-57% of the initial concentrations. At the start of the last renewal period, the actual measured test concentrations were 0.010, 0.021, 0.049, 0.096 and 0.21 mg/L in 4.6, 10, 22, 46 and 100% of the SS, respectively. At the end of this 24-hour renewal period these concentrations had decreased to 41-81% of the initial concentrations. Based on these results, the average exposure concentrations were calculated to be 0.010, 0.020, 0.046, 0.10 and 0.23 mg/L and used to determine the LC50 value.
