Short-term toxicity to aquatic invertebrates

Description of key information

In accordance to Regulation (EC) No. 1907/2006, column 2 of Annex VII chapter 9.1, a study on the acute toxicity to invertebrates does not need to be conducted if a long term aquatic toxicity study on invertebrates is available.

Additional information

One study investigating the short-term toxicity of Diisopropyl adipate (CAS 6938-94-9) to freshwater invertebrates is available. The non-GLP study was conducted under static conditions according to OECD guideline 202 using Daphnia magna as test organism (Bouillon, 2012). The test documentation is insufficient to assess the study fully (e.g. no analytical dose verification was performed, no information on the test concentrations given). The 48 h-EC50 was determined to be 31 mg/L based on nominal test concentration. Due to the lack of analytical monitoring, this test was considered to be invalid for assessing the toxicity of Diisopropyl adipate (CAS 6938-94-9) to freshwater invertebrates. Therefore, in accordance to Regulation (EC) No. 1907/2006 Annex XI, 1.5 Grouping of substances, a read-across to the structurally related category member Dibutyl adipate (CAS 105-99-7) was conducted. Further justification is given within the endpoint summary 6.1 and within the category justification section 13. In this case of read-across, the best suited (highest degree of structural similarity, nearest physico-chemical properties) read-across substance within this category was used.

The study with Dibutyl adipate (CAS 105-99-7) was conducted by a Japanese authority according to OECD guideline 202 using Daphnia magna as test organism (MOE, 1993). The test was only conducted for 24 hours which is considered as insufficient for assessing the acute toxicity to Daphnia magna. The 24 h-EC50 was determined to be 16.6 mg/L based on nominal test concentration. However, for this substance a valid GLP-guideline test according to OECD 211 with Daphnia magna is currently ongoing. Therefore, in accordance to Regulation (EC) No. 1907/2006, column 2 of Annex VII chapter 9.1, a study on the acute toxicity to invertebrates does not need to be conducted.
