Juvenile O. mykiss were exposed to 2 -butyl-2 -ethylpropanediol (BEPD) at nominal concentrations of 4.6, 10, 22, 46 and 100 mg/L in a semi-static 96 -hour acute toxicity test according to OECD 203 guidelines. The test design included an untreated control comprising of dilution water only. Concentrations of BEPD were verified analytically at test initiation and termination and mean measured concentrations were 92 to 119% of nominal. The 96 -hour LC50 determined was >100 mg BEPD/L (mean measured). The NOEC (no-observed effect concentration) was 23 mg BEPD/L (Bell, 1995).
The 48 hour acute toxicity of BEPD to Daphnia magna was carried out in a static system according to OECD 202 and EU guidelines. First instar daphnids were exposed over a 48 hour period to BEPD at nominal concentrations of 0 (control) and 100 mg/L. Duplicate, loosely covered vessels, each containing 10 daphnids in 200 ml medium were allocated to each treatment. Concentration of BEPD was verified analytically at test initiation and termination and mean measured concentration was 95 mg/L. Individual pH, temperature and dissolved oxygen values remained within acceptable limits throughout the duration of the study. The 48-hour EC50 was determined to be >100 mg BEPD/L (nominal) (Bell, 1995).
S. capricornutum was exposed to BEPD at nominal concentrations of 2.2, 4.6, 10, 22, 46 and 100 mg/L in a static 72 -hour toxicity test carried out according to OECD 201 guidelines. The test design included an untreated control comprising untreated algal growth medium only. Concentrations of BEPD were verified analytically at test initiation and termination and measured concentrations were 91 - 98% of nominal. The 72 -hour ErC50 and EbC50 endpoints determined were >100 and 94 mg BEPD/L, respectively. The corresponding 72 -hour NOEC based on inhibition of specific growth rate, determined using Williams' test analysis, was 45 mg BEPD/L (Bell, 1995).
Activated sludge obtained from a sewage treatment plant was exposed in a 3 -hour respiration inhibition test to concentrations of 0, 51.1, 103, 258, 515 and 2575 mg BEPD/L. The test design included duplicate samples and followed OECD 209 guidelines. The 3 -hour EC50 based on respiration inhibition was 650 mg BEPD/L (Solyom, 1998).
In the chemical safety assessment performed according to Article 14(3) in connection with Annex I section 3 (Environmental Hazard Assessment) no hazard was identified. Therefore according to REACH Annex I (5.0) exposure estimation is not necessary for long-term toxicity to fish and aquatic invertebrates. Consequently, in accordance with Column 2 of REACH Annex IX, the studies do not need to be conducted as all identified uses of the substance are assessed as safe for the environment.
