Measured values between Day 0 and Day 21 have a recovery of 86-100% for the testing range 125 to 1000 mg/L. As this concentration range is the relevant one for the derivation of the NOEC the nominal concentrations were used.
For 62.5 mg/L recovery was 41-95% but this concentration is not of relevance for the derivation of the NOEC reproduction. An explanation for the low recoveries at 62.5 mg/L might be that TAED hydrolyzes and biodegrades rapidly. - Details on test conditions:

- The NOEC and LOEC for the adult mortality were determined directly from the results. Significant deviations were determined in comparison to the control group using statistical standard procedures as Normality Test, Equal Variance Test and Analysis of Variance. For the determination of significant deviations for the reproduction rates and the body lengths One Way Analysis of Variance, p = 0.05 was used. One Way Analysis of Variance, DUNNETT’S Method, p = 0.05 was used for the determination of significant deviations for intrinsic rates of natural increase. Statistical evaluation of the age of first reproduction and the occurrence of stillborn juveniles and aborted eggs could not be carried out because Normality Test failed. The coefficients of variation around the mean number of living offspring produced per parent in the control and test groups were evaluated.

Intrinsic rate of natural increase:
The intrinsic rates of natural increase (IR) of the surviving parent animals accounting for generation time and offspring numbers were used for calculation of population growth and maintenance. The IR of the surviving daphnids of the test groups were compared to the control by One Way Analysis of Variance, Dunnett’s Method (p = 0.05).There was a statistical significance only at the concentration level 1000 mg/L when compared to the control, but no significant effect on the reproductive output.

T he Daphnia magna Reproduction Test (Semi-Static, 21 d) of Peractive P (batch No. DEA 4 TM 0014) was conducted according to OECD 211 (1998) from 2008-07-11 to 2008-08-01 at Dr.U.Noack-Laboratorien in Sarstedt, Germany, . Test system was Daphnia magna STRAUS (Clone 5). 10 test organisms, individually held, were used per concentration level and control. At test start they were 2 to 24 hours old. The test method was semi-static. The test solutions were renewed 3 times per week. Aim of the Daphnia Reproduction Test over 21 days was to assess effects on the reproduction capacity and other test item-related effects on parameters such as occurrence of aborted eggs and stillborn juveniles, time of production of first brood, adult mortality, intrinsic rate, dry body weight and length of the parent animals. Based on the results of the preliminary range finding test the study was performed with 5 test item concentrations ranging from 62.5 to 1000 mg/L set up in a geometrical series with a dilution factor of 2. The active ingredient Tetraacetylethylenediamine (TAED) and its main metabolite Diacethylethylenediamine (DAED) of all concentration levels and the control were analytically verified via HPLC of samples on days 3, 10, 17 (fresh media, 0 h) and on days 5, 12, 19 (old media, 48 h). The mean recoveries of the active ingredient TAED were in the range of 81 to 94 % of the nominal values in fresh and old media. Recoveries of the main metabolite DAED remained below LOQ in all samples. Analytical results are given in Table 23 and Table 24. All effect values are based on the nominal test item concentrations. In the Daphnia magna Reproduction Test with the test item Peractive Ponly significant effects on adult mortality occurred at the concentration level 1000 mg/L. Significant effects on reproductive output could not be observed.
· The adult mortality after 21 days amounted to 50 % at the concentration level 1000 mg/L. In the control as well as in the tested concentration levels 62.5 to 500 mg/L all daphnids survived until the end of the study. The EC50 for the adult mortality was determined directly from the results to be 1000 mg/L.
· The intrinsic rates of natural increase (IR) of the surviving parent animals accounting for generation time and number of offspring were used for calculation of population growth and maintenance.
· The average number of juveniles per parent in the control group was 133 after 21 days. The reproductive output was not statistically significant reduced at any of the tested concentrations.
· The coefficient of variation of the mean number of living offspring produced per parent in the control group was 11 %.
A summary of all test item related effects is given in Table 1. The effect levels (NOEC and LOEC) are given based on the adult mortality and intrinsic rates of natural increase as the most sensitive effects. The EC50 for the adult mortality was determined directly from the results.

· No stillborn juveniles and aborted eggs were produced by the control group. Related to the total number of produced juveniles (dead + alive) the percentage of dead juveniles was ≤ 1 % at all concentration levels.
· The first brood was released till day 9 in the control group and in the tested concentration levels 62.5 to 1000 mg/L by all daphnids.
· The mean body weight in the concentration levels 62.5 to 1000 mg/L ranged from 0.68 to 1.03 mg per daphnid. The mean body weight in the control was 0.85 mg per daphnid. The mean value of the body length in the tested concentrations was in the range of 5.05 to 5.45 mm per daphnid and 5.33 mm per daphnid in the control group.
Water quality parameters as pH-value, dissolved oxygen, water hardness and temperature were determined to be within the acceptable limits.
The EC50 of the reference item at 1.75 mg/L after 24 hours was within the prescribed concentration range of 1.0 - 2.5 mg/L of quality criteria according to AQS P 9/2 (05/1996) for daphnids clone 5 cultured in Elendt M4 medium. The EC50-value of the reference item is also within the recommended range of 0.6 - 2.1 mg/L according to OECD-Guideline 202.
