Description of key information

Aluminium has been shown to exhibit some degree of pro-oxidant activity [Exley 2004]. The results provided by [Dey 2008] identify SIRT1 as an AP-1modulator and suggest a novel mechanism by which alumina nanoparticles may function as a potential carcinogen.

Additional information

Inert, low toxicity, low solubility substances, such as Al2O3, when in the form of nanoparticles may promote an inflammatory response in epithelial cells, possibly due to production of reactive oxygen species (ROS) [Dey et al. 2008]. This response is considered an intrinsic property of nanoparticles as a result of their surface area as opposed to a response specific to the chemical substance.

The results from the in vitro studies are informative for mechanisms of action of possible relevance to the carcinogenicity endpoint. However, their relevance for assessing the carcinogenicity hazard in vivo is limited.
