The bioaccumulation and depuration of the test substance in rainbow trout was investigated by means of a fish feeding study in a dynamic flow through system (Burri, 2014). Biomagnification factors (BMF) in fish were determined. The fish were fed during 14 days in a flow-through system with food containing a target concentration of 1000 µg/g radiolabelled test item. After the exposure, the fish were transferred to flowing untreated water and the depuration of radioactivity was followed for further 28 days.

Temperature, pH and oxygen concentration during the whole study were within acceptable limits; measurements ranged from 13.3-14.2 °C, 7.8-8.2 and 8.4-9.5 mg/L, respectively. By determination of the radioactivity level, the content of the test item in the food was confirmed.

A level of 1053.5 µg/ g food was measured immediately after preparation. A level of 1038.3 µg/ g food was measured after 18 days of storage at -20 °C. At the end of the feeding period, the test item concentration in fish amounted to 8.523 ± 4.803 µg/ g fish. At day 1 of depuration, the concentration in the fish had decreased to 3.419 µg/ g fish. The level on day 7 was 1.622 µg/ g fish. At the end of the depuration phase (day 42), the radioactivity concentration was 1.249 µg/ g fish. An additional measurement was performed after 3 days of uptake. The mean value of 5 fish amounted to 14.487 ± 6.866 µg/ g fish.

Fish growth rates for the control fish and the test fish were similar. The growth rate of the test fish was 0.017 day-1. The rate constant k2 for the depuration of the radioactivity from the fish was calculated to be 0.051 day-1. Therefore, the growth-corrected rate constant k2g was 0.034 day-1. The growth corrected half-life was therefore 20.5 days.

The derived time 0 concentration C0,d was 3.122 µg/g and the content of test item in the diet was 1053.5 ng/g. Based on these values and a feeding rate of 0.02 g food/g fish/day, the substance assimilation efficiency alpha was calculated to be 0.0148.

Kinetic BMFK, growth-corrected BMFKg and lipid-corrected growth-corrected BMFKgL amounted to 0.0058, 0.0088 and 0.0427, respectively.
