PREPARATION AND APPLICATION OF TEST SOLUTION

- Method: A stock solution was prepared by adding 180 mg of the test substance to sterilised test medium and adjusting the volume to 1 L. The solution was stirred for 24 hours. The stock solution was diluted with sterilised growth medium to obtain the nominal test concentrations under sterile conditions: 7.7, 17, 37, 82 and 180 mg hexanol/L.

Calculation of the percent inhibition of growth rate, yield (cell counts) and biomass integral was performed according to the OECD guideline.
A concentration-effect relationship was observed and statistically analyzed to obtain effect concentrations.
The test results were statistically analyzed to determine EC10 and EC50 values together with 95 % confidence intervals using Probit-analysis assuming lognormal distribution of the values by using the computer program ToxRat.
The NOEC and LOEC values for yield and biomass were determined by the Welch t-test.

Above are results if the analytical monitoring of the test substance (1 -hexanol) over the experimental duration in all test concentrations with and without algae. The mean measured test concentrations varied in some cases by more than +/20% of the nominal values. 

The results are therefore interpreted with respect to the mean measured values. The values in bold are the values effectively used. All values at time 0 h were taken from the test solution prior to the addition of algae. Samples at time 48h and 72h were taken from the exposure vessels.

The acute toxicity of LOROL SPEZIAL to R. subcapitata was determined in sealed 72 h growth inhibition toxicity tests.
Algal inocula were exposed to test solutions which were not renewed during the test. The tests were repeated three times.

EL values are said not be accurately identified in many cases since there was large variation in the cell concentrations measured (this is not clear from the table of results reported, identical to Table 1 here). All of the concentrations tested in the unfiltered and filtered solutions were above the limit of solubility. Any effect seen at these concentrations should be taken with caution as they may represent effects deriving from physical fouling rather than true toxicity of the test substance. The inconsistent EC values and cell concentrations may well be explained by the fact that the concentrations tested are much higher than the limit of solubility.
